Spectrophotometer: a device that measures the amount of light travelling through a medium, and comparing it to the amount of light that travels through bacteria in the same medium. The denser the bacteria the less light will flow through. A sample can be taken to figure out how many viable colonies there are, so you can relate the number of bacteria present to the cloudiness. If you use a bacteria like lactobacillus, found in yoghurt, which requires b12 to grow, which means it can not produce it, but must get it from an external source. For it to grow, the medium must have vitamin b12. By using a nutrient rich medium (depending on bacteria) in this case put everything it needs to grow except b12, this causes the bacteria to not grow. Two extremes: lots of vitamin b12, or where b12 is not limiting factor.
