ELISA for secreted HBMS. Because HBMS is a secreted protein (46), we determined whether the HBMS protein produced in VSV - infected cells is secreted into the culture media. We detected HBMS in the media of VSV-HBMS infected cells but not in uninflected or recombinant WT VSV-infected cells by both western blot (data not shown) and qualitative ELISA. We found by quantitative ELISA that secreted HBMS levels reached 35 ng/ml in the media of VSV-HBMS infected BHK cells by 24 h post infection. Therefore, the HBMS produced in VSV- infected cells is correctly processed for secretion.
