Rab gtpases (monomeric) and rab effectors: docking & tethering vesicle, grabbing the vesicle. Snares (transmembrane proteins: fusing vesicle w/ target membrane. Rab gtpase recruited to vesicle- through rab gef. Rab interacts w/ rab effector- interacts to dock and tether. Specificity: many different rab gtpases & rab effectors. Where it"s going, and different compartments have different rab. 2 snares coil around one another, brings the vesicle tsnare- found on target membrane close to target membrane: helical domains- coil around each other- locks 2 membranes together. Specificity: different types of snares can"t bind w/ t- Coil so tightly/come so closely displace water molecules b/w membranes. Need to then separate/uncoil snare complex: nsf & adaptor proteins: unravel snare helical domains. Need to know structure of golgi apparatus (cis golgi network cis cisterna medial cisterna trans cisterna grans golgi network) Copii-coated vesicles bud from er exit sites: contains exit signals (soluble proteins & transmembrane proteins that is bound by cargo receptors)
