1. Mixed Inhibition
o It affects both Kcat and Km: as Kcat decreases
Km increases
o More common inhibitors
o Mixed inhibition makes it difficult for the
substrate to bind to the enzyme
Mixed: Km increased while Vmax decreases
- Non-competitive (mixed) inhibition: Both slope
and y-int increases with [I] but not necessarily to the
same extent
2. Uncompetitive Inhibition
o Inhibitor binds to ES complex
o Binding of the substrate may open up binding
site for the inhibitor
