Planting part of the sample on the bottom of the t-flask, then lay them down; need the bottom of the flask for bacteria to attach and grow on. Every culture is a little bit different no set time for growth. A routine 5 days or so is not enough. We use an inverted light microscope to look at these cultures. Cell attachment and growth are monitored with an inverted phase contrast microscope. We are looking for dividing cells, and when we see them we know we can harvest the culture. Tubulin is what forms the cytoskeleton of cells and the spindle fibers during mitosis. When spindle fibers form, it pulls tubulin from cytoskeleton from cell, which creates the round shape.
