Living cells (need to be capable of division) Not required but most of the time it is used to help prevent contamination: maintenance (time in the incubator; growing culture) Culture initiation most important requirement in cytogenetics: live cells capable of division are critical, how cells are collected and handled will influence cell growth and metaphase quality. Peripheral blood: drawn in sterile syringe or vacuum tube, preservative free, sodium heparin. This way, we are able to collect the lymphocytes: invert the tube to mix sample and anticoagulant. If we don"t invert to mix, it won"t work; if the tube is not inverted, it is the main reason things go wrong: sodium heparin has a green top but not all green tops are sodium heparin. Lithium heparin also have green tops and can be toxic to cells and should not be used.
