Lecture 9 - reading: signaling through enzyme-coupled receptors. Some tkr not activated by phosphorylation, but by conformational change by interaction between the two kinase domain outside the active site. Generate docking site for intracellular proteins (cid:314) large signaling complex. Intracellular signaling proteins have various structures, usually have pt-binding domain. Can also contain sh3: allow them to interact with other protein as part of the signaling process. 3 major ras proteins in human: h, k, n- ras. If ras is inhibited, the cell proliferation/differentiation process by rtk will not occur ras - gef: activate ras. Rtl either activate ras-gef or inhibit ras-gap (cid:314) activate ras. Grb2: adaptor protein links sev to sos. Sh2 domain binds receptor by recognizing phosphorylated tyrosine domain on it. The signal is short lived and it must be converted. Use: mitogen-activated protein kinase (map kinase module ) Ras-map-kinase signalling pathways are known as above. The activity period of map kinase depends on the kind of extracellular signal.
