Spread transformants on agar with x-gal: blue colonies (lacz intact no insert), white colonies (lacz gene disrupted insert present). Construction of cdna libraries: isolate mrna and convert to cdna before constructing a library; reverse transcriptase + oligo dt primer to synthesize 1st strand, degrade mrna with ribonuclease h and synthesize 2nd strand using dna polymerase. Libraries can be screened by complementation or hybridization with specific probes. Review topic 1. 2 techniques for dna / rna analysis. Electrophoresis: separation (based on size) through a gel matrix in an electric field. Dna is negatively charged and migrates towards the anode. Dna restriction mapping: create maps of dna based on size of restriction enzyme fragments. mapping requires the comparison of digestion patterns from different re used individually and in combination.
