Use of a mixture of enzymes to catalyze a series of reactions leading to formation of a color. The amount of color that develops is proportional to the amount of cholesterol or triacylglycerol you started with. Timing is not critical- simply need enough time for the assay to run to completion. Cholesterol ester + h20-> (cholesterol esterase) -> cholesterol + fatty acid. Cholesterol (oxidized) + o2 + h+ -> (cholesterol oxidase)-> cholest-4-en-3-one + hydrogen peroxide. Hydrogen peroxide combined w/ other compounds to produce a dye (quinoneimine dye) Triacylglycerols are hydrolyzed to glycerol and free fatty acids. Triacylglycerol + h20 -> (lipase) -> glycerol + fatty acid. Glycerol + atp -> (glycerol kinase) -> glycerol-3-phosphate + adp. Hydrogen peroxide combines with other compounds to produce a dye. Amount formed is proportional to amount of cholesterol that we started with.
