Note: a good task to assess function of hippocampus in rodents. Infusion of the nmda-r antagonist apv into ventricles blocks learning and memory in the morris spatial water maze task. This finding is consistent with the hypothesis that an ltp-like process is necessary for spatial learning. Functional nmda receptors require a glun1 subunit. Therefore, knocking out the glun1 gene effectively blocks the formation. Nmda receptors and memory formation: glun1 deletion of nmda receptors. Tonegawa"s group deleted the glun1 subunit in ca1. Ltp could be induced in the dentate gyrus (c) but not in the ca1 region (b). The ko mice showed impaired learning as compared to control mice, but did show some improvement in performance. Mice were unable to learn the location of the platform, even over many trials, as determined in the probe test (panel e) Much of what we know about ltp / memory is derived from blocking function.
