Iron-hematoxylin: best for looking at morphology (nucleus, etc. ) difficult to do, not done anymore, yellow/gray, trichrome stain, can be good if done well, blue background, purple internal structures, note: there will be a range of colors. If the stool passes out slowly, there"s more time for cyst formation: formed stool: more cysts, less trophozoites; higher cyst : trophozoite ratio, diarrhea: more trophozoites, less cysts; lower cyst : trophozoite ratio. The organisms: pathogenic amoeba, entamoeba histolytica, commensal amoeba, entamoeba coli, endolimax nana. Iodamoeba butschlii: entamoeba hartmanni, entamoeba polecki (bolded are the ones we need to know) May 7, 2020: other protozoan parasites, blastocystis hominis; often mistaken as an amoeba. Large karyosome, blot-looking : look for presence of peripheral chromatin (margination); may not always be seen, will appear as a defined ring of chromatin on periphery. If an invasive infection, look for trophozoites in tissue sections.
