Document Summary

In this lab we prepared experimental bacterial transformation using e. coli and a plasma dna to see acquired traits like ampicilin resistance acquired and fluorescence in the bacterial cells. Dna is transferred between bacteria using two methods- transformation and conjugation. In transformation bacteria takes up extra dna from its surrounding area. Conjugation requires contact directly between two bacterial cells. Frederick griffith first discovered transformation when he saw living cultures of normal streptococcus pneumonia were able to kill mice after being transformed with a heat killed pathogenic strain. In 1944 oswald avery purified rna, dna and proteins from s. pneumonia to see which one a very counted for transformation. Bacteria hold extra genes similar to their useful double stranded dna called plasmids, allow beneficial genes to be exchanged. Satellite colonies from untransformed cells grow around colonies. Recombinant dna technology has allowed us to connect genes from different sources to plasmids.
