Initiation begins with separate ribosomal subunits: firstly, the small 30s subunit binds to the initiation complex at the ribosome binding site, then the larger subunit joins to complete the ribosome, 3 initiation factors needed. If-3 stabilises 30s subunits binding and stops 50s subunit binding too early: also, checks accuracy of recognition of first trna, must be released before 50s subunit can join. If-2 binds initiator trna (formylated methionine) and controls entry to p site. If-1 binds near a site to prevent aminoacyl trnas entering too early.
