Wild type: accumulation of purified synthesized wt a-synuclein results in formation of aggregates ring like pore structures. Mutated: e57k is an artificial mutant of a-synuclein promotes formation of ring like pore structures. Wild type and mutated forms of a-synuclein were also able to form fibrils in different buffer, but partial form was not able to form fibrils. Overexpression of a-synuclein wt or mutated (variant) is toxic to dopaminergic neurons in snpc. Injection with wt or mutated in snpc formed fibers in brains. Injection of partial form did not form fibers in brain. Stain with tyrosine hydroxylase (histochemistry to mark for dopaminergic neurons) see loss of dopaminergic neurons in wt or mutated injected animals but not in partial form injected animals. Ring like structures is gain of toxic function.
