The association of ubiquitin and substrate protein is called ubiquination. Ubiquination regulates the expression of numerous cellular proteins. Unregulated cellular proteins are attributed to human diseases such as cancer. Deubiquitinases (dubs) reverse the ubiquination process by cleaving ubiquitin from the proteins. The degradation of some proteins are induced via the inhibition of dubs. This could potentially include cells that were previously seen as unaffected by pharmaceuticals. This article concerns itself with the use of highly specific dub inhibitors to inhibit ubiquitin specific protease 7 (usp7) to ultimately prevent or treat cancers. Ft671 destabilizes usp7 substrate like mdm2 -> increases levels of p53 -> transcription of p53 target genes -> induction of tumour suppressor p21 and inhibition of tumour growth. The inhibition of ubiquitin specific protease 7 (usp7) results in the degradation of oncogenic e3 ligase mdm2, and leads to the re-activation of the tumour suppressor p53 in various cancers. P53 tumour suppressor, lots of things lead to its degredation.
