After tissues have been fixed and embedded in wax they are thinly sliced and placed on glass microscope slides. The tissue is allowed to rest on the slides at an elevated temperature. To analyze the tissue, the wax is removed by dissolving it in an organic solvent. Since prior to analyzing tissue they were dehydrated, the tissue needs to be rehydrated for analysis. Grated serious of alcohol containing increasing concentrations of water is used to gently introduce water back into the tissues and proteins.
