Acquired fate in isolation = normal prospective fate as predicted by fate map. Part 2: heterotopic tissue transplantation (tissue grafting: put tissue in a place it does not belong. Grafted tissue forms a piece of neural tube on the ventral side: develops according to its former location (where it came from, neural fate is determined in late gastrula. Fate map versus specification map: do not know vegetal pole because cells do not differentiate in blastula stage, some tissues have been specified and some have not. Explant consisting of animal cap and vegetal region: (no separation by blastocoel) Ectoderm is separated from the endoderm by the blastocoel. Vegetal cells induce animal cap to develop mesoderm = conditional specification: the cells have to be able to respond to the signal, there is a certain window to which these cells are competent. Nieuwkoop centre: dorsal mesoderm induction: whole embryo only consists of 32 cells, used the dorsal side of the embryo.
