Willow bark has been used for thousands of years to make a tea as a treatment for pain, fevers, and in ammation. 1830s: the active ingredient, salicin, was isolated. 1860: it was realized that salicin could be converted into salicylic acid and that this resulted in a more effective treatment, but there are many side effects. 1890s: baeyer co. synthesized the acetyl derivative (acteyl salicylic acid - asa), which was effective and well tolerated: sold under the name aspirin. Difference is in the llers and in the price. Fillers can just cause small differences such as dissolving time, etc. Inhibits the formation of prostaglandins from arachidonic acid by inhibiting the cyclo-oxygenase enzyme. Knowing the asa mechanism allowed medicinal chemists to search for compounds that inhibit cox.
