Lecture 17 – February 10, 2017
MODULE OVERVIEW
Differentiation → effector cells (vector functions)
B cells – Activation
T Helper Cells – help to B cells/CD8+ T cells
Different roles for different T cells
- Activate B cells
- Activate macrophages to kill intracellular pathogens
- Directly kill infected cells
Egress to site of infection
- Efferent lymphatic vessels → thoracic duct → blood stream
Cell mediated immunity
- Th2 cytokines promote Tc Killing and Macrophage ROS killing
Humoral Immunity
- Abs promote pathogen neutralization, opsonisation, complement
Generation of BCR and TCR diversity
Downregulation of T cell activity and Immunological memory
Signal 3 – Differentiation
Three signals
- Review
- Signal one – pMHC:TCR
- Signal 2 – costimulatory glands
- Signal 3 – differentiation through cytokines
Lymphocyte activation = proliferation and differentiation
- Activated T cells
o Proliferate in response to IL-2
o Differentiate
▪ CD4+ T helper cells from THO to TH1 or TH2 or TFH or TH17 or others
▪ CD8+ T cells: from cytotoxic T lymphocyte (CTL) precursor to CTL
▪ Regulatory T cells

Document Summary
T helper cells help to b cells/cd8+ t cells. Th2 cytokines promote tc killing and macrophage ros killing. Downregulation of t cell activity and immunological memory. Effector t cells are the resulting differentiated t cells from activated t cells. They can be classified in the following types, depending on: the transcription factors they express, the profile of cytokines they make, their main function in adaptive immunity. Once activated, an effector t cells does not require costimulation to act. Answer: c: l-selectin is only expressed on na ve t cells and c is the only option where l-selectin is working with na ve t cells. Remember the name of 5 subtypes: th1 or th2 or tfh or th17 or treg, all secrete different cytokines. Differentiation of t helper cell subsets is regulated by polarizing cytokines. Apcs may bind pamps via prrs, inducing cytokine secretion.
