Methods: w/ transfer pipette place small drop of nigrosin near end of glass slide. W/ fresh transfer pipette, add drop of k. pneumoniae to nigrosine drop, use edge of a slide and spread drop into very thin smear varying from opaque black to gray (apply even pressure). Let smear dry flood smear w/ 1% crystal violet stain for 1 min then w/ transfer pipette flood slide with water (add from side) (this allows stain to float up and not precipitate on slide). Results: cells should appear pink to red while capsule should appear as a clear zone surrounding the cells, best place to examine capsule is where it"s not stained darkly. Method: add 500 ml distilled water to 1l flask.
