The dialysis bag containing the protein solution is placed in a buffer containing the desired final ingredients. Over time the concentrations of all molecules small enough to pass through the pore will equilibrate between both compartments, while the protein stays in the dialysis bag. Often several buffer changes are required to get the final concentration of the undesired compounds down to a minimal concentration. In the case of the anfinsen folding experiment, -me would initially be included in the dialysis buffer but the denaturant would not. Once the denaturant has been removed and the protein has refolded, the -me would then be removed. Know than many proteins are modified by being cleaved into 2 or more pieces, and that the term for this is proteolysis. Many proteins are cleaved or trimmed by cellular proteases after synthesis to an active form can be mechanism for regulating activity.
