Mouse model in jackson lab= obese mice & diabetic mice. 1950 discovery = recessive mutant mouse associated w/ massive obesity. Recessive defective gene = ob/ob obese recessively inherited form of obesity = db/db diabetes. Parabiosis = surgical union of 2 animals that leads to development of common circulation. 2 mice & fuse together so share same blood vessels/nutrients/hormones/proteins in blood. A- obese + wildtype = parabiotic unions of obese ob/ob mutants & normal lean littermates (+/+) Results = wildtype unchanged & obese lose weight. Share/give leptin to obese = decrease food. Intake & rapidly lost weight to normal level. B- diabetes + wildtype = parabiotic unions of diabetes db/db mutants & normal lean littermates (+/+) Results = diabetes unchanged & wildtype lose weight. Share/give leptin to wildtype (+/+) = decrease food.
