Document Summary

Reading #22- 12. 3, 13. 1, 13. 2- analyzing dna & genome annotation. Pcr allows a targeted region of a dna molecule to be replicated into as many copies as desired: selective and highly sensitive. Requires same basic components used by cell to replicate its dna. Need: template dna, dna polymerase, dntps, 2 primers. Oligonucleotides refer to the primer sequences; complementary to ends of region of dna to be amplified: (cid:1007)" p(cid:396)i(cid:373)e(cid:396) (cid:373)ust (cid:271)e oriented toward region to be amplified so that it creates a new complementary strand. Use dna polymerase that are heat-stable: taq polymerase: makes it so you do not have to add new dna polymerase each cycle. Need to test pcr products to see if it worked. Gel electrophoresis is a way to determine the size of dna fragments by inserting dna into wells in an agar-like solution and passing an electric current through it while submerged.
