An analog of colchicine (an extract of the seeds of the autumn crocus) Inhibits mitotic spindle formation by binding to tubulin. Prevents formation of the spindle fibers (made of tubulin) thus preventing separation of sister chromatids. Exposure is a tradeoff between quality and quantity. Mitogen usually used for peripheral blood, but not for bone marrow, amniotic fluid, and solid tissues. In us, trypsin/giemsa are most commonly used: swelling of cells w/ hypotonic solution. 20-25% fbs (fetal bovine serum) in distilled water. Mixtures of kcl and sodium citrate: fixation of cells w/ fixative. Must be prepared fresh before each use for optimal chromosome quality and staining.
