Exercise 1 pcr: looks at 16s rrna of mycoplasma to amplify. Uses experimental band (260bp), internal control (160bp, checks for internal problems - ex. no band = excess template), and ours did not have a sample band (570bp, can indicate insufficient primer or template source is not mammalian cell). Lateral gene transfer = transduction, conjugation or transformation. Transformation = direct transfer of naked dna, cells must be competent to do so (bacilus, streptococcus, haemophilus, neisseria). Are called transformants once they have received/taken up the dna. Antibiotic resistance is often one if the transferred genes (degrade antibiotic penicillin g or inactivate it by changing its structure streptomycin/chloramphenicol) often associated with transposons. E coli is not naturally competent, however it can be transformed by treating in cold with. Cacl2 or rbcl then heat shocking at 42 degrees for 1-2 min. Take up supercoiled dna more easily than relaxed circular or linear dna.
