Propylthiouracil (pt) inhibits synthesis of th by preventing organification and trapping of iodide to iodine/inhibition of coupling idootyrosines. Also inhibits peripheral conversion of t4 to t3. Methimazole inhibits synthesis of th by preventing organification of iodide to iodine & coupling idotyrosines. Decreases hr, myocardial contractility , blood pressure and myocardial oxygen demand. Its often necessary drug to control symptoms of thyroid storm.
