Neurons- sense changes in the environment, communicate changes to other neurons, command the body"s responses to these sensations. Size: cells: 0. 01-0. 005 mm in diameter= only seen after compound microscope was invented (1700s) Need thin slices of tissue- but didn"t have a device that would cut + not disturb the structure. Early 19th c solved the prob: fix tissues by putting them in formaldehyde + using microtome to make thin slices. Contrast: fresh brain has the same pigmentation, so can"t resolve individual cells. Stains take selectively colour some parts of cell in brain tissue= solved the problem. Dye stains the nuclei & material/ granules surrounding the nuclei (nissl body) = looks like a lump with a nucleus. Stains the cell body in particular (not everything else) Enables to study arrangement (cytocarchitecture) of neurons in different parts. Soaking brain tissue in silver chromate solution (golgi stain) some neurons would be darkened.
