Incubate in 37*c water bath for 10 minutes (amount of colour proportional to how much was original in the tube) Reverse pipetting: start with 2nd stop and end with only 1st stop (remaining volume back to original microfuge tube) Take absorbance readings at 415nm by pressing the green button.
