Being familiar with dilution calculations and the relationship between absorbance, path length, and concentration while understanding physiological function and regulation of glucose while determining its unknown concentration nad absorptivity. Prepare and mix 200ul (with blue set to 0200) prelab standards after labelling microfuge tubes with [glucose] & initials. 2 get 8 new microfuge tubes and label them 1-8 (see above) 6 get 16 vertical wells of a spectramax190 (duplicates) with 200ul (use blue set to 0200) Pipette 100ul of enzyme reagent in tubes 1-8 (with blue set to 1000) and add the samples (see above) Seal the tubes and mix by inverting a few times. Incubate in 37*c water bath for 10 minutes (amount of colour proportional to how much was original in the tube) Reverse pipetting: start with 2nd stop and end with only 1st stop (remaining volume back to original microfuge tube) Take absorbance readings at 415nm by pressing the green button.
