You needed very thin slices in order to view the tissue in the microscope but brain tissue is like jello and not easy to cut. To harden the brain tissue they immersed them in formaldehyde. To cut thin slices the microtome was invented. Stains were developed to stain some but not all parts of the tissus . Franz nissl showed that a class of dyes would stain the nuclei of cells and ask clumps of material surrounding the nuclei. Nissl stain useful for 2 reasons distinguishes neurons and glia from one another (stains only neurons) allows us to study the cytoarchitecture (arrangement) of neurons in different parts of the brain.
