Immediate early genes - encode for many proteins including transcription factor & growth factors. Neural activity/stimulation activates transcription of immediate early genes. They allow us to assay neural activity in fixed tissue. We stain the eigs to measure what areas were active prior to fixation then using. Ihc or ish we see expression of protein products. Often used as a comparison btw groups see how ieg expression varies. Assaying cellular function in fixed tissue & brdu. Use to examine cell proliferation & protein trafficking. Before fixing tissue, you introduce a marker while processes are occurring and then you detect it when examining the tissue. Brdu - synthetic analog of thymidine base gets incorporated into replicating. Dna indicates when a cell has divided visualized and detected with ihc. In dna synthesis phase of mitosis, cells can be marked by exposing them to tagged dna base pair. Dna base pair is tagged w/brdu or 3h-thymidine.
