Transformation procedure: freeze tubes with cells to prevent killing them with heat, add cells to prechilled tube. Add divalent cation to cells to associate to dna, neutralizing its charge, and disrupting the fluidity of its membrane: add dna sample to tube. Incubate on ice for 2 minutes (membrane potential depolarized).
