Dna footprinting is a method of investigating the sequence specificity of dna-binding proteins in vitro. This technique can be used to study protein-dna interactions both outside and within cells. The regulation of transcription has been studied extensively, and yet there is still much that is not known: how to find the start. Sigma factor is a part of initiation complex. Once initiation of rna transcription is complete, the sigma factor can leave the complex. Only one dna strand is transcribed (template strand); newly transcribed mrna is identical to non- template strand. Only a fraction of the genome is transcribed, while in dna replication, all of the dna is replicated. Different enzyme with different properties: transcription -- rna polymerase; dna replication -- dna polymerase ( much faster, more accurate). Slightly different base: u instead of t, ntps instead of dntps. Both extend the nucleic acid chains in 5"-3" direction.
