The purpose of this lab is to demonstrate the complexity of lipids found in different types of tissues. This will be achieved by obtaining endoplasmic reticulum (er) rich and mitochondrial fractions from calf liver tissue and separating them using phase chromatography. This will be followed by analyzing the er fraction using 2-d thin layer chromatography, as well as performing lipid and protein assays using the mitochondrial fraction. the lipids and protein absorbance assays will be performed using 559 nm and. Tlc plate with corresponding labels for all of the lipids. G plate was labeled and activated (heated at 100 c for 1-2 hours). Firstly, the plate was placed in a tank containing the basic solvent system: chcl3-meoh-ammonia (65:35:5 v/v). Then, the plate was turned 90 degrees and placed in in the second tank with the acidic solvent system: chcl3-acetone-meoh-acetic acid-h2o (10:4:2:2:1; v/v). Lastly, the plate was dried and placed in an iodine saturated tank for 5 minutes.
