NEU 365D Lecture Notes - Lecture 16: Acetaldehyde Dehydrogenase, Aldehyde Dehydrogenase, Alcohol Dehydrogenase
Document Summary
Pharmacogenetics the study of the influence of heredity on the responses to drugs, or their fates in the body (pharmaco- dynamic & kinetic aspects) When administering a drug, a physician would like to be able to predict, explain and control variability in the responses of drugs. Ex: sometimes the dose is too high/low in comparison to general dose for the population. Metabolism disorders that leads to a buildup of substrates that normally would be metabolized by non-functioning enzymes issues. Will show the symptoms typical for the disorder. Pharmacogenetic defect/variant tend to be silent" in the absence of drug challenge. Nobody knows about them until there is a drug given to the patient and it becomes evident. Ex: alcohol asian flush not a good predictor who has it until you"ve tested it. Alcohol-dehydrogenase to acetaldehyde is normally kept really low in our bodies. Normally acrylaldehyde is rapidly metabolized by aldehyde dehydrogenase acetic acid.
