Assume plasma and all tissues in which the drug equilibrates constitute a single compartment. Single rate constant - removal of drug. Different rates of distribution to different tissues. Each compartment has different rates of distribution for the drug entering and leaving it. Equilibration rates - dependent on blood profusion in the tissue type as well as the concentration gradient, p m / b value, etc. No metabolism/excretion from second compartment (small amount but negligible) K value = metabolism from liver & excretion from kidney. Equilibration of drug from blood to tissue elimination. Equilibration dependent on blood profusion in the tissue type & concentration gradient. Driving force drug into tissue until they are equal. Ex: drug concentration introduced as i. v. at a location. Measure [drug] in blood and tissue (i. e. brain) Concentration in blood always lower than tissue. By the time the blood leaves the tissue (equilibration has occurred - they were the same)
