Guinea pig ileum (part of the intestine) - common subject for dose-response study. Hang up ileum and measure force of contraction with application of various drugs. Minimum threshold dose where the drug starts to work. Log scale used to be able to distinguish doses (over a wide range of doses and maintain accuracy) Use straight-ish middle part for statistical analysis. Compare different drugs that may be acting at the same sites (expect same types of responses) Same effect produced by different drugs that act by identical or similar mechanisms. Find approximal dose at which you get a half-max response. At higher doses - maximally-effective doses due to saturation. Limited number of binding sites for drug to bind and illicit its response. Potency - amount needed to achieve the desired effect. More potent - smaller dose needed to achieve desired effect (left-ward shift) Potency - not equated to binding affinity (can be influenced by it)
