GENE 3200 Lecture Notes - Lecture 4: Complementary Dna, Plasmid, Knockout Mouse
Document Summary
In mice with brown fur, (bb) stem cells contributed to germ line: b chromosome could be either cftr(-) or cftr(+, test for cftr(-) via pcr. If gene of interest is a recessive lethal, you are screwed you will only find heterozygous. Can"t create a knockout mouse with a recessive lethal**** Take some of cells and inject them into a mouse embryo. (done at blastocyst phage) this is being done in a black mouse blastocyst (bb). Cells from original donor are bb and the cells we are injecting are bb. We are implanting these embryos into a surrogate female, so we can let them grow and develop and examine the offspring. Offspring are chimeric (modeled black/brown because some of cells from stem cell donor (injected cells) and other cells come from original embryo (black)) We use coat color to keep track of our construct injected.
