Make a proper aseptic smear for each of the 4 cultures. On a good nutrient agar plate, streak escherichia coli culture and label the plate. Place inoculated tubes and plates in incubation at. 37 c for e. coli and 30 c for s. cerevisiae. Heat fix smears and stain with crystal violet for every culture in step 1. Using a small stream of water, rinse the slides at an angle so the water does not hit the smear directly, rinse until the water on the smear itself is nearly colorless. Blot excess liquid from slide using bibulous paper- do not rub the slide on the paper. (paper can be discarded in regular trash) Examine the smears of 2 bacteria at 1000x - draw them. Streak sabouraud dextrose agar (sab) slant (yellow cap) from the saccharomyces culture with inoculating needle - one single streak up center of slant.
