BSCI 223 Lecture Notes - Lecture 11: Covalent Bond, Microscope, Hydrogen Bond
Document Summary
Incineration: heating loops to red hot will kill everything, nothing can survive. Effects of high temperatures (above maximum temperature: denature proteins. Once unfolded, will not refold back into original structure. High enough heat will undo the dna helix: interfere with integrity of membranes and cell wall, disrupt structure and function of nucleic acids. Thermal death point: lowest temperature that kills all cells in broth in 10 minutes: when trying to sterilize something, consider the thermal death point. Thermal death time (tdt): time to sterilize volume of liquid at set temperature. Decimal reduction time: time to achieve 10x reduction in number over a given time: amount of time it takes to kill 90% of the organism. At optimal temperature, growth will be steep. At minimum temperature, growth will be less steep. Used to disinfect, sanitize, sterilize, and pasteurize. More effective than dry heat: water is great at transferring heat, air is not. Methods of microbial control using moist heat: boiling, autoclaving.
