09/23/2015 (cid:1) (cid:1) (cid:1) (cid:1) (cid:1) (cid:1) (cid:1) (cid:1) (cid:1) Botulism: 4 forms of disease all lead to flaccid muscle paralysis. 1-3 days after consumption of food, patient becomes weak and dizzy. Abdominal pain, blurred vision, fixed dilated pupils, dry mouth. If food isn"t cooked thoroughly and is placed in an anaerobic environment, the bacteria mature and neurotoxin is formed (cid:1) (cid:1) (cid:1) (cid:1) (cid:1) (cid:1) Mortality reduced to 5-10% currently via supportive care. Spores germinate and colonize the intestinal tract- an infection. Period before toxin effects are seen is longer. The toxin can be concentrated for aerosolization. Single polypeptide- cleaved to a and b fragments (light & heavy chains) that remain connected by disulfide bond. Toxin is complexed w/ proteins to protect it while in intestinal tract. Binds receptors on motor neurons which stimulates endocytosis of the toxin. Acidification of the endosome releases the light chain which inactivates proteins that regulate acetylcholine release (required for excitation)
