As you have seen, nucleic acids are synthesized beginning at the 5" ( five prime ) end and progressing towards the 3" ( three prime") end. Here"s one experiment performed by early investigators: early investigators did not know what the structure of dna was, or how it could be a gene. They just knew it was a long string of nucleotides (a, t, g, c). They thought it was a boring repeat block copolymer : atgcatgcatgcatgc : this is what her diffraction pattern of the dna looked like, a young american postdoc named jim watson was working in a cambridge.
