Because it"s selective doesn"t mean it won"t affect/inhibit others (increased concentrations that claim to be selective start to hit other subtypes in that receptor class) Concentration dependent: bad if people take more than they should. Not intended to have effects on other parts, but depending on dose, concentration, and metabolism, can have unintended effects. Drugs: viagra, verdanifil, tadalafil (cialis, gated ion channels - gaba (inhibitory), glutamate (excitatory), nicotinic acetylcholine (excitatory, ligand + voltage gated ion channels. Ligand: gaba, nicotinic ach, glutamate: membranes are electrically polarized. The inside of the cell is typically negative compared to the outside (-50 - - The membrane potential is largely due to electrogenic na+, k+ atpase. This establishes the membrane potential because ions tend to move down their electrochemical gradient across the polarized membrane.
