Lecture 7: transcription: rna poly unwinds dna, does not proofread and correct mistakes. This is why we have mistakes in proteins: reads template 3" 5", but builds 5" 3", adds nucleotides. Pulls the rna poly, destablizes the dna/rna interactions: may have some of it in the transcript, but most of it is not included in transcript. Transcription of a eukaryotic gene: prokaryotes don"t have modifications to their mrna, only eukaryotes, 5" cap is added while transcription is still happening. Modified gtp, added right on the +1 site: 3" poly(a) tail: added where the termination sequence would have been, extend the length of the mrna transcript. About 100-300 adenines added: mrna transcripts that have a longer polya tail are more stable in the cytoplasm and will have a longer life.
