Most habitats contain microbes, so its necessary to separate them. To maintain and keep track of such small subjects, we have to grow them under artificial conditions. Microbes are everywhere and its very easy that your sample may become contaminated causing misleading results. Sample is placed into a container of sterile medium containing appropriate nutrients to sustain growth. Spreading the sample on the surface of a solid medium, or into a flask or tube. O2 and co2 may be required for the growth of certain microbes. When is it considered to be a culture? observable growth that appears on the medium after incubation. Incubation: placing the sample into an incubator creating the proper growth temperature. Isolation: isolated microbes may take the form of separate colonies on solid media. Further isolation requires taking a bit of growth from an isolated colony, and moving it to another medium (1 way to make a pure culture)
