When a signal come in it causes the molecule to go through a conformational change which causes the gdp falls around (lower surplus than gtp in the cell) Then the gtp binds again to another molecule which activates the molecule sending the signal out. Gtp is a lot like atp it can go through an exergonic hydrolysis reaction. Meaning the reaction turns itself off by having the phosphate just randomly turn itself off. Needing another signal to turn it back on can reactivate if the ligand is still bound. A mutation in an individulas genome (dna) causes a g-protein to bind gtp even when it is not receiving a signal as part of a signal transduction pathway. It is inactive and then a phosphate is added (very negative high charge) whether the amino acid is positive or negative determines where it moves toward the negative phosphate or away.
