C4f6: antibody to misfolded which targets epitope for domain in protein that is hidden when the protein is folded correctly but becomes exposed when the protein is folded correctly. The first one shows the anterograde transport: rate of transport. In c when they took out the misfolded, the phenotype was rescued. The misfolded causes change in transport: the native gel (do not need to denature proteins which allows it to see the native conformation) This was to study c4f6 and how it binds to sod1. C4f6 only tags sod1 ox but not wt sod1 in native gel. In sds-page, it tags both because they are linearized which makes all domains accessible: fals- can have different mutants including ones that are not sod1. The disease does not directly depend on sod1 mutation and misfolding: effects of ox-sod1 on bcl-2 is the same as sod1 and this associates with mitochondrial dysfunction.
