Tetrahydrofolate absorption: folate polyglutamate is unstable b/c hydrolyzed by enzymes in stomach & gi tract. It"s found in diet (unstable when cooked or stored) Hydrolysis is zinc dependent: folate monoglutamate is compound that"s transported into cells. Metabolized in intestinal lumen: 5-methyl thf is an important source of 1c for exchange good for rapid growing cells (gut/bone marrow) 5-methyl thf is taken up by liver & bone marrow via atp dependent transporter. 1c entity removed & transferred to homocysteine forming methionine. Homocysteine +1c methionine; 5-methyl thf thf: thf can be used in cell or stored by adding glutamates to it, resynthesizing as polyglutamate. Thf is the active form of folic acid. Generation of folate derivatives: one way to add 1c to thf is by adding formyl group to it through formate to form 10 formyl thf. Thf + formate 10 formyl thf: uptake of meoh is bad- a bit as 10ml can make you blind & 30ml can kill you.
