Per1/per2/cry1/cry2 proteins form a complex that inhibits the clock-bmal1 complex from activating the transcription of the 5 target genes. Rev-erb inhibits bmal1 transcription: ck1 phosphorylates and degrades the per proteins in the cytoplasm. The cry proteins can then bind to that ck1-per complex and prevent further degradation go to nucleus to cause inhibition: fbxl3 f-box protein degrade cry1/2 in cytoplasm.
