BIOL 4310 Lecture Notes - Lecture 17: Advanced Sleep Phase Disorder, Circadian Clock, Arntl
Document Summary
Lecture 17 march 17 circadian clocks and the genetics of sleep disorders. Also allows us to help individuals not getting enough sleep. Circadian clock = internal self-sustaining oscillator that organizes our bodies activities around a 24 hour day (incl sleep-wake cycle, which can be affected if our clock is misaligned) Process s - sleep debt, increase during activity (subjective day) and decreases during sleep. Dep on time of onset, may sleep for more or less time. Ttfl: clock + bmal1 form complex that bind e-box and activate transcription of per1, per2, cry1, cry2 and rev-erb . Per1/per2/cry1/cry2 proteins form a complex that inhibits the clock-bmal1 complex from activating the transcription of the 5 target genes. Rev-erb inhibits bmal1 transcription: ck1 phosphorylates and degrades the per proteins in the cytoplasm. The cry proteins can then bind to that ck1-per complex and prevent further degradation go to nucleus to cause inhibition: fbxl3 f-box protein degrade cry1/2 in cytoplasm.
