Exp 1: showed mutation to c. elegans ortholog pi3k increased life span. Pi3k is responsible for activating akt more cell proliferation. Reduced also the fertility after the mutation. Exp2: mutation daf-2 caused a dramatic extension in life. Daf-2 is the insulin receptor daf-16 is a down stream factor transcription factor similar to foxo in mammals which regulates transcription in the absence of pi3k. Pi3k removes foxo by akt from the nucleus. Also removing daf-16 would lower the life span. Slide 6: orthologs in mammals, daf-2 age1/pi3k pdk1 akt inhibits daf-16/foxo. The health of the organism is higher when daf-16 is active thus this is done by having less insulin or substrate for daf-2. It controls self-renewal: a pathway that activates the self-renewal and accumulation of these cells.
