Electrons pass through and stain areas that are dark. Scanning em you can"t see inside of the cell. Gives a 3-d image surface only need to coat specimen"s whole surface with heavy metals. Can only look at outside of the object. When looking at membrane we use transmission em using thin slice. Funcion of membrane: barrier for water soluble molecules you don"t want them to go through the membrane, separates outside and the inside of the cell. Organelles vs. cytosol (inside the cell: maintain ion gradients, signalling funcions plasma membrane receptors. But there is way more lipid (but it weights lighter than protein) Amphipathic molecule: have hydrophilic head + hydrophobic tail.
